fill it up to 50 psi or so, stick it in a pool or stationary tub and look for bubbles. if its the valve stem, you have to get a new one put in and clean/ seal the rim. you cant do this yourself without a tire machine. (same deal with a bead leak) if its an object in the tread more than an inch away from the sidewall, you can pull it with some pliers and use a patch kit that is sold at pep boys, autozone ect.
keep in mind the only way to patch a tire when its still on the rim is a pull thru patch and is not an ideal method.
